Abstract. Interactions between soluble proteins in the cell nucleus lead to formation of discrete spherical droplets, known as nuclear organelles, which are vital for cellular regulation. However, the physico-chemical properties of the nuclear organelles are still poorly understood, because they are highly challenging to study in a live cell environment, without disruption of their biological functions. Recently, developments of two complimentary optical techniques, - Raman spectroscopy and Fluorescence Lifetime Imaging (FLIM), made possible to investigate the formation, and maintenance the nuclear organelles. These techniques have unraveled that the molecular content of the nuclear organelles is dynamically changed during the cell development and can also serve as a marker of abnormal cellular states. This presentation will focus on the recent developments of Raman Spectroscopy and FLIM techniques for studies of the cellular function.
